Lympho24+SpinMediumisareadytouse,sterilemediumforpreparationofPeripheralBloodMononuclearCell(PBMC)from12-48houroldperipheralblood,buffycoat,bonemarrow,orcordbloodbyasimplecentrifugationprocedure.ItreducestheRBCcontaminationbutalsoreducesPBMCyieldby20%.

MostdensitygrADIentmediaarelimitedfortheuseoffreshwholeblood.Lympho24+SpinMediumisrecommendedfortheuseofwholebloodthatisolderthan8hours.Thishelpstoreducethetimedependedcontaminationwithgranulocytes.

Theenrichedmononuclearcellscanbeusedforawiderangeofdownstreamapplicationssuchasmagneticactivatedcellsorting(MACS¹)orstemcellresearch.

SeparationschemeforPBMCenrichmentwithLympho24+SpinMedium

AtfirstcarefullylayerthesamplematerialontopoftheLympho24+SpinMedium.Avoidamixingofthetwophases.Alternativelyit’spossIBLetousepluriMatetubes.Thebarrierpreventsamixingofsamplematerialanddensitysolution.FurThermorethetubeenablespouringofftheenrichedcellsafterthefirstcentrifugationstep.Afterthedensitycentrifugationaspiratetheupperlayer(Plasmaanddilutionbuffer).Afterwardstransferthemononuclearcelllayertoanewconicaltubeandwashthecells.
